A PhD position is available at the Department of Food Science and Technology (Food Biotechnology), University of Natural Resources and Life Sciences (BOKU). Our project aims at an in-depth functional characterization of the Glucose-Methanol-Choline (GMC) family of flavin-dependent oxidoreductases (Auxiliary Activities Family 3) of the mycoparasitic fungus Clonostachys rosea. This gene family is significantly expanded in the genome of this fungus, but the biological role of the encoded enzymes as well as the reason for the presence of such a high number of encoding genes is poorly understood.

The main tasks of the project include establishment of a CRISPR/Cas9 system for single/multiple inactivation of GMC oxidoreductase genes in C. rosea, heterologous protein expression and biochemical characterization of GMC oxidoreductases that encode functional enzymes, and screening for GMC oxidoreducatase enzymes that possess direct and indirect antimicrobial activities that could be applied for the control of fungal/bacterial plant diseases.

Experimental  methods comprise, among others, aseptic work with fungal cultures, standard molecular biological techniques such as nucleic acid extraction, PCR, gene cloning/plasmid construction, expression analysis via immunoblot, qPCR, as well as CRISPR/Cas9 genome editing, heterologous expression of proteins in a suitable (eukaryotic or prokaryotic) host, protein purification and biochemical characterization of the expressed proteins.

The position is funded by the FWF for the duration of 3 years. We provide an inspiring and supportive work and social atmosphere, supervision by expert scientists as well as access to modern state of the art equipment.

As an optimal candidate, you should hold a master’s degree in Biochemistry, Molecular Biology, Biotechnology or similar, be highly motivated and proactive, have an analytical mind and enjoy scientific research. Team spirit, being at ease with independent work and very good oral and written English skills are expected, experience in project-relevant methods will be an asset.
